RESUMO

Acetaminophen (AC) is one of the most popular pharmacologically active substances used as an analgesic and antipyretic drug. Herein, a new type of hollow Fe3O4-rGO/GCE electrode was prepared for electrochemical detection of AC through a three-step approach involving a solvothermal method for the synthesis of hollow Fe3O4 and the chemical reduction of graphene oxide (GO) for reduced graphene oxide (rGO) and Fe3O4-rGO nanocomposites modified on the glassy carbon electrode (GCE) surface. The as-prepared Fe3O4-rGO nanocomposites were characterized using a transmission electron microscope (TEM), X-ray diffraction (XRD), and a magnetic measurement system (SQUID-VSM). The magnetic Fe3O4-rGO/GCE electrodes were employed for the electrochemical detection of AC using cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and square wave voltammetry (SWV) and exhibited an ultra-high selectivity and accuracy, a low detection limit of 0.11 µmol/L with a wider linear range from 5 × 10-7 to 10-4 mol/L, and high recovery between 100.52% and 101.43%. The obtained Fe3O4-rGO-modified GCE displays great practical significance for the detection of AC in drug analysis.

Transcription factors of the SOX family were first discovered in mammals in 1990. The sex-determining region Y box 9 belongs to the SOX transcription factor family. It plays an important role in inducing tissue and cell morphogenesis, survival, and many developmental processes. Furthermore, it has been shown to be an oncogene in many tumors. Gynecological malignancies are tumors that occur in the female reproductive system and seriously threaten the lives of patients. Common gynecological malignancies include ovarian cancer, cervical cancer, and endometrial cancer. So far, the molecular mechanisms related to the incidence and development of gynecological malignancies remain unclear. This makes it particularly important to discover their common causative molecule and thus provide an effective therapeutic target. In recent years, studies have found that multiple mechanisms are involved in regulating the expression of the sex-determining region Y box 9, leading to the occurrence and development of gynecological malignancies. In this review, we discuss the prognostic value of SOX9 expression and the potential of targeting SOX9 for gynecological malignancy treatment. We also discuss progress regarding the role of SOX9 in gynecological malignancy pathogenesis through its mediation of important mechanisms, including tumor initiation and proliferation, apoptosis, migration, invasion, chemoresistance, and stem cell maintenance.

Fluorescent carbon dots (CDs) were hydrothermally synthesized from a mixture of frozen tofu, ethylenediamine and phosphoric acid in an efficient 64% yield. The resulting CDs exhibit good water solubility, low cytotoxicity, high stability, and excellent biocompatibility. The CDs selectively and sensitively detect Co2+ through fluorescent quenching with a detection limit of 58 nM. Fluorescence can be restored through the introduction of EDTA, and this phenomenon can be used to quantify EDTA in solution with a detection limit of 98 nM. As both analytes are detected by the same CD platform, this is an "off-on" fluorescence sensor for Co2+ and EDTA. The technique's robustness for real-world samples was illustrated by quantifying cobalt in tap water and EDTA in contact lens solution. The CDs were also evaluated for in vivo imaging as they show low cytotoxicity and excellent cellular uptake. In a zebrafish model, the CDs are rapidly adsorbed from the intestine into the liver, and are essentially cleared from the body in 24 h with no appreciable bioaccumulation. Their simple and efficient synthesis, combined with excellent physical and chemical performance, renders these CDs attractive candidates for theranostic applications in targeted "smart" drug delivery and bioimaging.

The sensitive and selective detection of endogenous glutathione (GSH) is of great significance due to its key role in physiological and pathological processes. Herein, we report a novel on-off-on transferrin (Tf)-coated gold nanocluster (AuNCs@Tf) with low toxicity, good stability and intense red fluorescence which can enter living cells and is mainly located in lysosomes. In addition, the fluorescence of AuNCs@Tf is quenched by Cu2+, indicating its sensitivity to Cu2+ with a detection limit of 0.23 µM at pH 4.5. Interestingly, we found that the quenched AuNCs@Tf-Cu2+ system only has a special affinity for GSH with a detection limit of 2.86 µM so that invisible AuNCs@Tf-Cu2+ can be gradually illuminated by endogenous GSH in the lysosomal environment of tumor cells with increase in time. More excitingly, the AuNCs@Tf-Cu2+ system can illuminate tumor cells rather than normal cells, resulting in its successful application in cancer cell recognition, which implies its great potential application in cancer diagnosis. Moreover, the AuNCs@Tf was also applied in Cu2+ strips and encryption/decryption of information.

We report a controllable strategy for fabrication of green and blue fluorescent carbon nanodots (CDs), and demonstrate their applications for pH and Cu(2+) sensing in living cells. Green and blue fluorescent CDs have been synthesized by hydrothermal method and pyrolysis of leeks, respectively, providing an easy way for the production of CDs without the request of tedious synthetic methodology or the use of toxic/expensive solvents and starting materials. Green fluorescent CDs (G-CDs) exhibit high tolerance to pH values and external cations. Blue fluorescent CDs (B-CDs) can be applied to pH and Cu(2+) sensing. The linear range of Cu(2+) detection is 0.01-10.00 µM and the detection limit is 0.05 µM. For pH detection, there is a good linearity in the pH range of 3.5-10.0. The linear and rapid response of B-CDs to Cu(2+) and pH is valuable for Cu(2+) and pH sensing in living cells. Confocal fluorescent imaging of human cervical carcinoma cells indicates that B-CDs could visualize Cu(2+) and pH fluctuations in living cells with negligible autofluorescence.

We report a facile and eco-friendly strategy for the fabrication of green fluorescent carbon nanodots (CDs), and demonstrate their applications for bio-imaging, patterning, and staining. A one-pot hydrothermal method using various plant petals yields bright green-emitting CDs, providing an easy way for the production of green fluorescent CDs without the need for a tedious synthetic methodology or the use of toxic/expensive solvents and starting materials. The as-prepared CDs show small size distribution and excellent dispersibility. Their strong green fluorescence is observed when the excitation wavelength is between 430 nm and 490 nm. Moreover, they exhibit high tolerance to various external conditions, such as pH values, external cations, and continuous excitation. Due to minimum toxicity as well as good photoluminescence properties, these CDs can be applied to in vitro and in vivo imaging, patterning, and staining. According to confocal fluorescence imaging of human uterine cervical squamous cell carcinoma cells, CDs penetrate into the cell and enter the cytoplasm and the nucleus. More strikingly, carp is directly fed with CDs for in vivo imaging and shows bright green fluorescence at an excitation wavelength of 470 nm. In addition, the obtained CDs are used as fluorescent inks for drawing luminescence patterns. Finally, we also apply the CDs as a fluorescent dye. Interestingly, the absorbent filter paper with staining emits dramatic fluorescence under 470 nm excitation.
